Publisher SummaryDipyridamole is an adenosine reuptake inhibitor and a phosphodiesterase inhibitor with antiplatelet and vasodilating activity. One of these is through the inhibition of phosphodiesterase enzyme in platelets, resulting in an increase in intraplatelet cyclic AMP and the consequent potentiation of the platelet inhibiting actions of prostacyclin. Another is the direct stimulation of the release of this eicosanoid by vascular endothelium, and the third is the inhibition of cellular uptake and metabolism of adenosine (thereby increasing its concentration at the platelet vascular interface). The chapter outlines the procedure for synthesizing of dipyridamole. The chapter discusses the physical characteristics, including ionization constant, solubility characteristics, x-ray powder diffraction pattern, thermal methods of analysis, ultraviolet spectroscopy, vibrational spectroscopy, fluorescence spectrum, nuclear magnetic resonance spectrometry, mass spectrometry. The chapter presents the several methods for the analysis of dipyridamole. Dipyridamole is highly bound to plasma proteins and is metabolized in the liver, and is mainly excreted as glucuronide conjugates in the bile. Excretion may be delayed by enterohepatic recirculation.
