Fine structural analysis of the localization of acetylcholinesterase accessible to intraventricular ambenonium chloride (cas 115-79-7) in rabbit brain

The light and electron histochemical distribution of cerebral acetylcholinesterase activity, protected in vivo by intraventricular administration of the highly specific, lipid-insoluble, reversible acetylcholinesterase-inhibitor ambenonium chloride (cas 115-79-7) was examined in rabbits by the enzyme protection-enzyme inhibition method of Koelle (1957). Ambenonium chloride can only gain access to the acetylcholinesterase extracellularly located along the plasma membrane, leaving the intracellular enzyme activity unprotected against a lipid-soluble irreversible enzyme inhibitor administered later. The distribution of the protected sites, indicating the intracerebral passage of the ambenonium chloride, was exactly the same as that of sites known as accessible to other extracellularly spreading materials.The distribution of the reaction end-product and the diffusion behaviour of ambenonium chloride suggest that the sites of acetylcholinesterase that are protected by ambenonium occur in the carbohydrate-containing outer layer of the cell membrane and in the extracellular substance.
