Publisher SummaryIn the presence of inorganic phosphate, guanosine is converted to guanine and ribose-1-phosphate by nucleoside phosphorylase. Guanase converts guanine to xanthine, and xanthine oxidase converts xanthine to uric acid; the extinction at 293 nm increases. After the addition of uricase, uric acid is converted to allantoin; the extinction at 293 nm decreases accordingly. The decrease of the uric acid concentration, as measured by the decrease in extinction at 293 nm, is a measure of the amount of guanosine in the assay system. This chapter describes a method that is used for the determination of guanosine in pure aqueous solutions. Nucleoside phosphorylase is not specific for guanosine; it also hydrolyses inosine, adenosine, and other derivatives, for example, the corresponding deoxy derivatives. Xanthine oxidase oxidizes xanthine, hypoxanthine, and purine. In contrast, guanase and uricase are very specific for guanine and uric acid respectively. The assay is also possible in the presence of guanylic acid, providing the system does not contain alkaline phosphatase.
