Oxyfedrine (cas 15687-41-9), a new aminoketone of the phenylethylamine series, was studied in three cardiac preparations in the cat. Oxyfedrine (0.5 × 10−6 g/ml) exerted a positive inotropic effect while 5.0 × 10−5 g/ml exerted a negative inotropic effect in the cat papillary muscle preparation. Positive inotropic, and coronary vasodilator effects were elicited by oxyfedrine in the cat Langendorff preparation. However, premature ventricular systoles and first degree heart blocks appeared with the two highest concentrations (10−7 g/ml and 10−6 g/ml). In the intact cat, oxyfedrine initiated positive inotropic and chronotropic effects as well as as marked decline in mean arterial blood pressure due to systematic arterial vasodilation. Oxyfedrine could not be demonstrated to be an independent coronary vasodilator. These findings indicate that oxyfedrine may not be of significance in the therapeutics of shock.
