Cocaine-elicited behavior and toxicity in dogs pre-treated with methiothepin or p-chlorophenylalanine

SummaryMethiothepin and p-chlorophenylalanine prolong and intensify the initial ataxia and prostration produced by a large intravenous dose (5 mg/kg) of cocaine in dogs. Subsequent behavioral, motor, and toxic effects of cocaine are completely blocked by methiothepin, and completely or partly blocked by p-chlorophenylalanine. It appears that serotonergic mechanisms function both in adaptation to the initial ataxia and prostration produced by intravenous cocaine, and in mediation of its behavioral, motor, and toxic effects. These mechanisms may be activated in large part by release of presynaptic stores of serotonin.
