Determination of the deacylation rate of p-anisoyl plasminogen streptokinase activator complex (APSAC, EMINASE) in human plasma, blood and plasma clots☆

The hydrolysis of the p-anisoyl group from the active centre of the thrombolytic proenzyme APSAC has been studied in human plasma, whole blood and thrombin-induced plasma clots using a radiochemical method based on a tritiated acyl group and in human plasma by a fibrinolytic method which exploited the inertness of acyl enzymes towards plasma inhibitors. In human plasma and blood, a half-life for deacylation of approximately 105–120 min independent of agent concentration, was found. The deacylation half-life was not significantly altered by the presence of human fibrin in either lysing or stable plasma clots nor was it changed in the presence of specific human antistreptokinase antibody
