  • The occurrence of D-3-phosphoglycerate dehydrogenase in animal tissues

    1.1. d-3-Phosphoglycerate: NAD oxidoreductase (d-3-phosphoglycerate dehydrogenase) has been partially purified from chicken liver.2.2. The distribution of this enzyme has been compared to that of d-glycerate dehydrogenase (d-glycerate:NAD(P) oxidoreductase). A marked variation in the ratio of phosphoglycerate to glycerate activity was observed from one animal to another. The highest levels of d-3-phosphoglycerate dehydrogenase were found in birds. The absence of significant amounts of this enzyme in rat-liver preparations requires a reinterpretation of the results of Ichihara and Greenberg on the pathway of serine biosynthesis in this animal.3.3. The product of oxidation of d-3-phosphoglycerate was identified as phosphohydroxypyruvate by preparation and characterization of the 2,4-dinitrophenylhydrazone of the enzymic product.4.4. The oxidation of NADH2 in the presence of phosphohydroxypyruvate and enzyme affords a convenient method of assay for this keto-acid.5.5. The product of enzymic reduction of phosphohydroxypyruvate and to be d-3-phosphoglycerate by chromatography on Dowex-1 chloride, followed by assay with colorimetric, enzymic, and polarimetric methods.
