Publisher SummaryThis chapter discusses analogs involving the replacement of the sulfur of pantetheine, 4'-phosphopantetheine, and coenzyme A with other atoms. In recent years, several analogs of coenzyme A and of some of its component building blocks have been synthesized. Presumably, antimetabolites of this type should be capable of providing information about the relative importance and the roles of the various parts of coenzyme A in the biological reactions in which it participates. Coenzyme A plays a crucial role in group transfer and energy transfer reactions. The role of the single sulfur atom of coenzyme A in transferring acyl groups or in activating the methylene group adjacent to the thioacyl carbon of esters of coenzyme A for condensation reactions can be visualized fairly well in terms of organic reaction mechanisms. Pantetheine represents a major portion of coenzyme A. It is required for the growth of certain microorganisms such as Lactobacillus helveticus and will replace coenzyme A, at least partially, in certain systems such as the adenosine triphosphate (ATP)-dependent acetylation of 4-aminoazobenzene by an extract of pigeon liver acetone powder.
