Dienol heptafluorobutyrates as derivatives for gas liquid chromatography of steroidal Δ4-3-ketones

The reaction of testosterone-17β-acetate (3) with heptafluorobutyric anhydride in high purity and anhydrous acetone at room temperature leads to the preferential formation of 1 with less than 1% of the isomeric 2,4-diene. The reaction is pseudo-unimolecular and after 30 min, 99% of the starting material is esterified. The two isomers have been fully characterized (UV, IR, NMR, CD, MS) and their sensitivities to electron capture detection are compared.
