Nephrotoxicity and hepatotoxicity of tris(2,3-dibromopropyl)phosphate in the rat☆

The nephrocarcinogen TRIS(2,3-DIBROMOPROPYL)PHOSPHATE (cas 126-72-7) (Tris-BP) caused extensive acute renal tubular necrosis at doses of 175 mg/kg, ip, and higher in rats. Pretreatment with cobalt chloride lead to a moderate reduction in Tris-BP nephrotoxicity, whereas pretreatments with the cytochrome P-450 inducers phenobarbital or polychlorinated biphenyls were without marked effects. Tris-BP administration also resulted in hepatotoxicity, but this effect was less pronounced and occurred at higher doses compared to Tris-BP nephrotoxicity. Tris-BP depleted glutathione levels both in kidneys and in liver, but pretreatment with diethyl maleate did not alter Tris-BP kidney necrosis. Monobrominated Tris-BP analogs showed very little kidney toxicity.
