Conversion of tryptophan to indole-3-acetic acid and other metabolites by Ceratocystis fagacearum☆

In a medium containing tryptophan, Ceratocystis fagacearum synthesized an auxin characterized as indole-3-acetic acid (IAA) by chromatography, bioassay, mass spectrometry and chemical derivatization. This synthesis paralleled fungal growth during 15 days in still culture, about 0·3% of the available tryptophan being converted to IAA. Intermediates between tryptophan and IAA were not detected in culture filtrates or mycelial extracts. Other products of an indole pathway for tryptophan metabolism were identified as tryptophol, O-acetyltryptophol and indole-3-aldehyde. The fungus also degraded tryptophan by an aromatic pathway yielding kynurenine, anthranilic acid and 3-OH anthranilic acid. These findings are discussed in relation to the stimulation of growth processes in infected oaks.
