Peroxidase-based biocatalysis in a two-phase system for allylic oxidation of α-pinene

Green synthesis of verbenol was achieved via allylic oxidation of α-pinene with a peroxide agent in the presence of a peroxidase enzyme in a biphasic aqueous-organic system. The aqueous part contained the enzyme together with the oxidation agent, while the organic part was the reservoir of α-pinene and hosted the reaction products. This design afforded a highly selective separation of the reaction products. Screening of the peroxidase enzymes indicated UPO mutant (PaDa-I) as the most efficient biocatalyst for this process. The optimization of the experimental conditions considered the composition of the two phases and reaction conditions (e.g. the peroxidase enzyme, the oxidation reagent and its concentration, the organic solvent and reaction time). Following this, a maximal conversion of 68% α-pinene and 44% yield in verbenol were achieved over 24 h. Very important, the aqueous phase containing the enzyme was possible to be reused for five reaction cycles after refreshing the content of oxidation reagent.
