The acute toxicity of amidephrine mesylate was examined in mice, rats, rabbits, dogs, monkeys and cats and compared with that of phenylephrine hydrochloride in mice. Amidephrine was less toxic than phenylephrine, and with both agents the effects were those of adrenergic stimulation. Amidephrine was singularly low in irritation potential and caused no pathologic changes following intranasal administration to rats and rabbits. Chronic po administration to rats and monkeys produced no discernible adverse effects and, in a two year study, no alteration of tumor incidence in rats.
