Acetyl-dl-phenylalanine β-naphthyl esterase activity in human muscle disease

Human skeletal muscle homogenates were found to contain an esterase that catalyzes the hydrolysis at neutral pH of acetyl-dl-phenylalanine β-naphthyl ester, a known low molecular weight substrate for chymotrypsin-like enzymes. The muscle enzyme was inhibited by phenylmethyl sulfonyl fluoride but not by ethylenediamine tetraacetate and p-hydroxymercuribenzoate. When the levels in patients were compared with controls, significant increases in muscle esterase levels were observed in patients with muscular dystrophies, polymyositis, and certain denervating diseases. These results suggest the possibility that this esterase may be involved in the muscle wasting process in human muscle diseases.Serum acetyl-dl-phenylalanine β-naphthyl esterase levels were normal in patients with Duchenne dystrophy and polymyositis.
