DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) A single i.p. injection of 12-O-tetradecanoylphorbol-13-acetate (I)(TPA) [16561-29-8] induced a transient increase in the levels of rat liver putrescine [110-60-1], spermidine [124-20-9], and spermine [71-44-3]. These polyamine concns. continuously increased until 6 h, immediately following administration of the tumor promoter. Phenobarbital (PB) [50-06-6] induced an increase of the putrescine and spermidine concns. during the 8 h post administration studied, while spermine reached a plateau after 4 h. When retinyl-acetate (RA) [127-47-9] was injected 1 h prior to TPA, the increases of the polyamines were considerably inhibited. This treatment with RA partially inhibited further increase of putrescine and spermine by PB. The findings are discussed in respect to the concomitant ornithine decarboxylase [9024-60-6] activities. .

Retinol [68-26-8] had little effect. Human erythroid stem cells were also stimulated by 13-cis-retinoic acid (max. at 10-7M) and esp. I (max. at 10-8M), but not by retinol. I also inhibited the growth of human myeloid leukemia cell lines KG-1 and HL-60, but not human K562 or mouse M1 cell lines. The mechanisms for these activities of retinoids are discussed. .
