Herein, an electrochemical method was developed to deter-
mine the activity of α-fucosidase (AFU) in serum samples
based on the quantification of p-nitrophenol (pNP) with a
screen-printed carbon electrode. It was found that the opti-

fication. This method was found to be suitable for measur-
ing a minimum serum AFU activity of 2.1 U/L and
achieved a recovery range of 97–110%. With the homemade
electrochemical mini-cell, only 0.5 ml of solution, ie. 50 μl
of serum sample was required to determine the AFU activ-
ity. Overall, cyclic voltammetry was demonstrated to pro-
vide better measurement accuracy than absorption
measurements. This electrochemical method was also
more economical than fluorescence spectrometric methods
and immunoassays. The electroactive compounds that exist
in blood samples, such as vitamin C, urea, uric acid, tyro-
sine, and glucose, did not interfere with the R2 signal. The
developed electrochemical method therefore exhibits con-
siderable potential for monitoring the AFU activity at
point-of-care sites for the diagnosis of liver cancer.
