Chemical structures of cocaine and other tropane-based DAT inhibitors
Structure-Activity Relationship Studies on a Series of 3α-[Bis(4-fluorophenyl)methoxy]tropanes and 3α-[Bis(4-fluorophenyl)methylamino]tropanes As Novel Atypical Dopamine Transporter (DAT) Inhibitors for the Treatment of Cocaine Use Disorders
Figure 1
Cocaine (1, Figure 1) is a psychostimulant and a highly addictive drug of abuse. In 2015, an estimated 1.8 million people aged 12 and older were current cocaine users. However, to date, no medication to treat cocaine abuse has proven successful.Cocaine binds to the dopamine transporter (DAT) and inhibits the reuptake of dopamine from the synapse into presynaptic neurons, resulting in an increase in extracellular dopamine levels.
