6-Demethyltetracycline, when heated with nitric acid, forms a stable colored derivative having an absorption maximum at 425 mμ. 6-Demethylchlortetracycline under the same condition is unstable. When 6-demethylchlortetracycline is heated with nitric acid, the color produced by the 6-demethyltetracycline fraction can be measured directly or extracted with n-butanol.
