Publisher SummaryThis chapter discusses the physical properties and synthesis of propoxyphene hydrochloride. Propoxyphene hydrochloride is a white, crystalline powder with no noticeable odor and a bitter taste. The melting range is 162.5–168.5°C. The specific rotation for a one percent solution should be between +52° and +57°. Propoxyphene hydrochloride has a unique characteristic in that the specific rotation value has an apparent dependency upon the concentration of the solution being measured. The original synthesis is of the α-dl mixture. A solution of benzylmagnesium chloride is added to a solution of α-methyl-8-dimethylaminopropiophenone and this solution is refluxed after one hour. The reaction mixture is decomposed with saturated aqueous ammonium chloride. The ether solution containing the 1, 2-diphenyl-2-hydroxy-3-methyl-4-dimethylaminobutane is decanted and then treated to form the hydrochloride. A comprehensive study of the crystallography of propoxyphene hydrochloride is documented in the chapter. Crystallization from ethyl acetate results in orthorhombic prisms elongated parallel to the b axis. The NMR spectrum of propoxyphene hydrochloride is also elaborated in the chapter.
