A comparative analgesia study of PROPOXYPHENE HYDROCHLORIDE (cas 1639-60-7), propoxyphene napsylate, and placebo

A total of 342 patients with postoperative or fracture pain were divided into 7 groups and were treated with one of the following: 32, 65, or 130 mg of propoxyphene hydrochloride; 50, 100, or 200 mg of propoxyphene napsylate; or placebo. Each of the treatment groups experienced significantly more analgesia than did the placebo group, with the exception of those patients receiving propoxyphene hydrochloride, 32 mg. Parallel doseresponse curves were obtained for propoxyphene hydrochloride and propoxyphene napsylate. Estimates of relative potency, based on several measures of subjective response and calculated on the amount of propoxyphene base in each salt, revealed that propoxyphene napsylate is 1.25–1.49 times more effective than propoxyphene hydrochloride, even though the milligrams of propoxyphene base are equivalent at each drug level.
