Formulation, in vitro and in vivo evaluation of ferrous sulfate loaded microemulsions

In this study, novel nonionic microemulsions (MEs) stabilized using a mixture of Span 20 and Tween 80 were developed for the transdermal delivery of ferrous sulfate. The MEs were evaluated in terms of their droplet size and zeta potential as well as their rheological properties using bob and cup rheometer. In addition, the flux of ferrous sulfate loaded in the MEs through shaved rat's skin was studied using Franz diffusion cells. The in vivo efficacy of developed MEs was evaluated in anemic rats over two weeks. The droplet sizes of the developed MEs were less than 180 nm and decreased with increasing ferrous sulfate content. The microemulsions showed a Newtonian viscosity. Ferrous sulfate's flux was proportional to ferrous sulfate concentration with a maximum flux achieved being 13.03 μg/cm2*hr. Furthermore, the treated induced anemic rats with the developed microemulsion relieved rapidly in comparison to received microemulsion free ferrous sulfate rats. Finally, the developed microemulsions demonstrated efficient carrier for transdermal application of ferrous sulfate.
