Precursors of glycine in the nervous system: Comparison of specific activities in glycine and other amino acids after administration of [U-14C]glucose, [3,4-14C]glucose, [1-14C]glucose, [U-14C]serine or [1,5-14C]citrate to the rat

A study has been conducted to measure the transfer of label from [1-14C]glucose, [3,4-14C]glucose and [1,5-14C]citrate to glycine, serine and other metabolically active amino acids in the central nervous system of the rat.Our results indicate that in vivo carbon atoms for the synthesis of glycine are derived primarily, if not exclusively, via a pathway leading through serine. Within the confines of the present experiments, no evidence was obtained to permit the conclusion that the glyoxylate pathway contributes significantly to the synthesis of glycine in vivo.The patterns of14C-labeled amino acids formed after the injection of [14C]glucose and [14C]citrate are in agreement with the concept of multiple metabolic pools for amino acids and tricar☐ylic acid cycle intermediates in the central nervous system. Intracisternally administered [1,5-14C]citrate is metabolized within the metabolic pool which selectively labels glutamine over glutamate.The contribution of the pentose shunt to glucose metabolism in the central nervous system appears to be reflected by the accumulation of label in GABA after the injection of [3,4-14C]glucose.
