Chemical stability of the potent bacterial mutagen (Z)-2-chloro-3-(dichloromethyl)-4-oxobutenoic acid (MX) and some related compounds at simulated Ames test conditions

The stabilities of the strong mutagen (Z)-2-chloro-3-(dichloromethyl)-4-oxobutenoic acid (MX), its geometric E-isomer (E-MX), and its methyl ether 3-chloro-4-(dichloromethyl)-5-methoxy-2(5H)-furanone (MeMX) were studied at simulated Ames test conditions. MeMX was found to hydrolyse completely to MX within five hours at 37°C. Simultaneously, the MX formed was partly isomerized to E-MX, and further hydrolysed to a highly hydrophilic product. These results, together with mutagenicity data obtained for MeMX and MX, indicate that the extremely high mutagenic activity previously reported in the literature for MeMX, at least partly might have been generated by MX formed during the incubation time of 48 hours used in the Ames test.
