Molecular Mechanisms of Parathyroid Hormone Type 1 Receptor Signaling

The parathyroid hormone (PTH) receptor type 1 (PTHR) is a G protein-coupled receptor (GPCR) that transmits PTH and PTH-related peptide (PTHrP) signals into cells. While acting via the same receptor, PTH and PTHrP trigger distinct physiological processes: endocrine regulation of bone turnover and mineral ions metabolism for PTH, and paracrine development of bone, heart, mammary glands, and other tissues for PTHrP. This article summarizes the current molecular, structural, and signaling basis responsible for the functional differences between PTH and PTHrP.
