The determination of thiols and of total glutathione in human blood using 6,6′-dithiodinicotinic acid (CPDS)

Two spectrophotometric methods are described for the rapid and accurate determination of thiols and of total glutathione in human blood. Both methods rely on the conversion of 6,6′-dithiodinicotinic acid by thiols to 6-mercaptonicotinic acid. The methods permit: (a) the specific determination of thiols at levels of 1 μg SH (equivalent to 10 μg glutathione), and (b) the specific determination of total glutathione at levels of 0.3 μg. The latter method is based on a specific enzymatic cycling reaction of glutathione. Both determinations may be performed rapidly and easily using a standard spectrophotometer.
