The tritiated dihydroartemisinine (I) [71939-50-9] produced was differentially accumulated from low concns. in culture medium into erythrocytes infected with P. falciparum. Uninfected erythrocytes concd. the drug less than 2-fold, whereas infected erythrocytes achieved more than 300 times the medium concn. The uptake process is reversible and saturable, with a dissocn. const. at the hypothetical receptor of 10.5 nmol/L. Competition studies indicate that the receptor is the same as that for artemether [71963-77-4], another quinghaosu deriv. Chloroquine [54-05-7] showed an interesting partial inhibition of uptake but was unable to release the bound radiolabeled drug from infected cells.
