  • (CAS NO.: ), with its systematic name of 1H-1,4-Benzodiazepine-3-carboxylic acid, 7-chloro-5-(2-fluorophenyl)-2,3-dihydro-2-oxo-, ethyl ester, could be produced through many synthetic methods.

    Following is one of the synthesis routes:

    Synthesis of Ethyl loflazepate

    2-Amino-5-chloro-2-fluorobenzophenone (I) is condensed with monoethyl methoxycarbonyl aminomalonate (II) and the crude product obtained (III) is deprotected by HBr to produce the free amine hydrobromide (IV). This product is then cyclized in a mixture of acetic acid-sodium acetate to afford CM-6912, which is separated by filtration and recrystallized in ethanol.
