Treatment of 2'-deoxy-3',5'-bis-O-<(1,1-dimethylethyl)dimethylsilyl>-uridine with sec-BuLi in the presence of TMEDA resulted in a highly regioselective lithiation at C-5.The lithiated derivative was treated with a number of electrophiles to afford 5-substituted 2'-deoxyuridines.
