The synthesis of a fluorescein-derived fluorogenic substrate, D,L-myo- inositol 1-([6'-hexyloxy-3-oxo]spiro[isobenzofuran-1(3H),9'-[9H]xanthen]-3'- yl hydrogen phosphate) sodium salt 9, for assay of activity of phosphatidylinositide-specific phospholipase C (PI-PLC), is described. This substrate is shown to be a sensitive substrate for bacterial PI-PLC in a continuous, non-radioactive assay.
