The roots of I. pubescens were purchased from Bozhou,
Anhui province and identified by Professor Jincai Lu,
Department of Pharmaceutical Botany, School of Traditional
Chinese Materia Medica, Shenyang Pharmaceutical Univer-
sity. A voucher specimen (AP-20120601) was deposited in
the herbarium of the Department of Natural Products Chem-
istry, Shenyang Pharmaceutical University.

Extraction and isolation
Acid hydrolysis of compounds
The dried roots of I. pubescens(15 kg) were powered and
reꢁux extracted with 70% EtOH (90 L × 2 h) three times.
The ꢀltrate was concentrated and yielded the crude sam-
ple extract after removing the solvent under vacuum. The
obtained crude extract was suspended in water (15L) and
then partitioned successively with petroleum ether, EtOAc,
and n-BuOH (15L × 3). The n-BuOH extract was evapo-
rated under reduced pressure to obtain a residue (300 g),
which was subjected to macroporous resin eluting with

M HCl (5 ml) and heated under reꢁux for 5 h at 90 ℃ with
