In conclusion, we have developed a selective
nitration protocol for the preparation of C-7 nitrated
8-aminoquinolines using easily installable and
removable ancillary groups. Mild and metal-free
reaction conditions, ambient temperature, use of tert-
butyl nitrite as nitrating agent, wide functional group
tolerance are the notable advantages of our
methodology. Experimental results suggest that the
reaction likely proceeds through a radical pathway.
We believe the present operationally simple selective
nitration strategy will gain much importance in
organic synthesis, medicinal chemistry as well as in
material sciences.

N-(Quinolin-8-yl)benzamide (1a) (0.2 mmol, 49.6 mg) was
taken in acetonitrile (2 mL) in a reaction tube and stirred at
room temperature for few seconds. TBN (3.0 equiv., 71.3
µL) was added to it and stirred at room temperature for 8 h.
After completion of the reaction (TLC), the reaction
mixture was quenched with water (2 mL). The reaction
mixture was then extracted with ethyl acetate. The organic
phase was dried over anhydrous Na2SO4 and concentrated
under reduced pressure to get the crude residue which was
purified by column chromatography on silica gel (60-120
mesh) using petroleum ether:ethyl acetate = 4:1as an eluent
to afford the pure product (2a) (44 mg, 76%) as a white
solid.
