Publisher SummaryCoenzyme A is a constituent of practically every living cell. It is the coenzyme for the transfer of acetyl groups and for lipid metabolism. It takes part in a large number of important biochemical reactions. Some of those are suitable for the determination of CoA. In the catalytic assays, CoA is reformed in the course of the reaction so that the rate of the reaction is the measure of the CoA content. Those methods are generally very sensitive. This chapter describes two assays for the determination of CoA-SH and CoA-S-S-CoA. The assay with 3-hydroxyacyl-CoA dehydrogenase (HOADH), according to the end-point method, measures CoASH, dephospho-CoA, pantetheine, N-(acetyl-β-alanyl)-cysteamine, N-acetyl-cysteamine, and other compounds that can act as biochemical precursors of CoA. That method can be used as a universal test for all potential CoA-active compounds. The assay can be so arranged that oxidized compounds and acyl-CoA can also be determined. The assay with phosphotransacetylase (PTA) only determines intact CoA. The rate of the reaction with dephospho-CoA is negligible. When the PTA assay is used as an end-point method, interference can occur from Na+, phosphate, and acetyl-Co. The chapter also discusses the catalytic assay and the fluorimetric assay.
