Synthesis of 1-O-alkylethylene glycol phosphonic acid analogues of lecithin and CEPHALIN (cas 1405-71-6)

The synthesis of dodecylethylene glycol-2-(2-trimethylammoniumethyl)phosphonate and dodecylethylene glycol-2-(2-aminoethyl)phosphonate is described. The ethylene glycol dodecyl ether phosphonate analogues of lecithin and cephalin were obtained by the direct esterification of ethylene glycol dodecyl ether with throughly dried 2-bromoethyl phosphonic acid, using tri-isopropylbenzene sulphonyl chloride (TPS) and pyridine as the condensing agent. From the intermediate 1-O-alkylethylene glycol-(2-bromoethyl) phosphonate, the direct phosphono analogues of lecithin and cephalin were obtained by direct amination procedures. The resulting phosphonates were characterized by elemental analyses, thin-layer chromatography (TLC) and IR spectroscopy.
