Antidepressant agents. I. Chemistry and pharmacology of amino substituted spiro[5H dibenzo[a,d]cycloheptene 5,1' cycloalkanes]

A series of spiro compounds structurally related to the common tricyclic antidepressants has been tested as inhibitors of the neuronal reuptake of noradrenaline and 5 hydroxytryptamine. Also, some behavioral tests have been performed. Two of the substances, N,N dimethylspiro [5H dibenzo[a,d]cycloheptene 5,1' cyclohex 2' en] 4' amine and N,N dimethylspiro [5H dibenzo[a,d]cycloheptene 5,1' cyclohexan] 4' amine, are very active in the NA uptake inhibition assay. A discussion on structure activity relationships is given.
