Some new N,S-substituted 2-nitrodiene compounds from reactions of mono(p-fluoroarylthio)-substituted dienes with amines

2-Nitrodiene compound 1 was stirred with p-fluorothiophenol for a long time and compound 3 was obtained. Compound 1 gave bis(thio)substituted 2-nitrodiene compound 4 and tris(thio)substituted compound 5 with 2 moles of p-fluorothiophenol in the presence of NaOH in ethanol. The compounds 9a-g have been prepared from 8a-g and 3. Compound 7 was obtained from the reaction of mono(thio)substituted 2-nitrodiene with morpholine. Compound 3 gives 11a-d in the reaction with piperidines in CH₂Cl₂ (or ether). Compound 13a-b have been obtained from the reaction of compound 3 with primary amines 12a-b. Compound 3 gives 15 and 16 in the reaction with 2,5-dimethylpiperazine in CH₂Cl₂.

2-Nitrodiene compound 1 was stirred with p-fluorothiophenol for
a long time and compound 3 was obtained. Compound 1 gave
bis(thio)substituted 2-nitrodiene compound 4 and tris(thio)substituted
compound 5 with 2 moles of p-fluorothiophenol in the presence of NaOH
in ethanol. The compounds 9a–g have been prepared from 8a–g and 3.
Compound 7 was obtained from the reaction of mono(thio)substituted
2-nitrodiene with morpholine. Compound 3 gives 11a–d in the reac-
tion with piperidines in CH2Cl2 (or ether). Compound 13a–b have
been obtained from the reaction of compound 3 with primary amines
12a–b. Compound 3 gives 15 and 16 in the reaction with 2,5-
dimethylpiperazine in CH2Cl2.

Durch die Reaktionen der 2-Nitrodiene mit Thiolen wurden mono-,
bis- und tetrakis(thio)-substituierte Dienverbindungen erhalten. Die
Reaktionen von 2-Nitrodiene mit S-, S,S-, N,S-, N,N- und O,S-
Nucleophilen haben cyclische Thioether wie Dithiole, Dithian-,
Oxathiolan-, Thiazol- und Benzthiazolverbindungen geliefert.1–6
Es ist bekant, daβ die Reaktionen von Polychlorbutadien- und
Polychlorbuten-verbindungen mit manchen Thiolen und Dithiolen thio-
substituierte Verbindungen mit Butenin- und Butadienstrukturen
liefern.7–12
Dem Unterstu¨tzungsfond fu¨r die Forschung der Istanbuler Universita¨t sprechen wir

Wir haben in dieser Arbeit neue Verbindungen durch die Reaktionen
von Mono(arylthio)substituierten 2-Nitrodienen mit Piperidin,
Piperidinderivaten, Piperazin, Piperazinderivaten und Aminen
dargestellt und charakterisiert.
Die C C
Bindung der Nitrovinylgruppe in der Nitrodien-
verbindung 1 hat wegen des negativen Induktiveffekts eine starke elek-
trophile Wirkung.
Nach langem Ru¨hren der 2-Nitrodienverbindung
1 mit p-
Fluorthiophenol entsteht die Mono(arylthio)butadienverbindung 3
unter Abspaltung von HCl. Diese Reaktionen sind langsam. Die Reak-
tion der Dienverbindung 1 mit zwei Mol Thiol liefert in wa¨βrigem
Ethanol in Gegenwart von Natronlauge die Verbindung 4. Die
Verbindung 3 mit zwei Mol p-F-Thiophenol liefert die Verbindung 5
(Schema 1).
Wahrscheinlich entstehen diese Verbindungen durch einen Addition-
Elimination-mechanismus. Die Verbindungen 3, 4 und 5 sind stabile,
gelbfarbige, kristalline Diene.
Monoaryl- und Diarylpiperazinverbindungen sind in der klinischen
Chemie wichtig und in einigen Medikamenten verwendet wor-
den. Finige Piperazinderivate sind beim Gen-Transport benutzt
worden.13–16 Wir haben vor kurzen einige Reaktionen von mono
(alkylthio-/arylthio-)substituierten 2-Nitrohalodienen mit Piperazin
und Piperidin untersucht.17–19
Die neue Verbindung 11d zeigt im IR-Spektrum eine HO- Bande
bei 3000 cm−1. 3 liefert mit 2,5-Dimethylpiperazin (14) in CH2Cl2
die Verbindungen 15 und 16. Die Verbindung 15 zeigt im IR-
Spektrum eine ❍✟NH Bande bei 1550 cm−1 und im–1H-NMR-
Spektrum bei ca. δ = 2.5–2.8 ppm fu¨r die ❍✟NH
Gruppe ein
Multiplett.
Durch die Reaktionen der Verbindung 3 in CH2Cl2 mit den
Verbindungen 8a–g sind neue S,N-substituierte Dienverbindungen 9a–
g erhalten worden. Die neue Verbindung 9e zeigt im IR-Spektrum eine
charakteristische ❍✟C O bande bei 1700 cm−1
.
Aus 3 entstehen in CH2Cl2 mit den Verbindungen 10a–d die
Verbindungen 11a–d und mit 12a–b die Verbindungen 13a–b als
stabile, gelbfarbige N,S-substituierte Verbindungen. Die Reaktio-
nen der Verbindung 3 mit N-Nukleophilen laufen wahrschein-
lich durch einen Addition-Eliminationsvorgang ohne Katalysator
bei Raumtemparatur ab. Die Strukturen dieser Verbindungen
sind durch Elementaranalysen und spektroskopischen Methoden
besta¨tigt.

Die H-NMR-Spektren der Verbindungen 13a–b zeigen bei ca. δ =
11.4 ppm fu¨r die HN Gruppe ein Singlett.

VG-ZAB-SPEC-Spektrometer. Schmelzpunkte (nicht korrigiert): Bes-
timmung in Kapilaren (Apparat Bu¨chi 510). Elemantaranalysen: Carlo
Erba 1106 und Elemental-Analyser. Du¨nnschichtchromatographic:
Merck DC-Alufolien Kieselgel 60F. Sa¨ulenchromatographic: Kieselgel
der Korngro¨sse 0.063–0.20 mm (Fa Merck). Der verwendete. Petro-
lether siedete bei 40–60◦C. Alle Lo¨sungsmittel wurden destilliert.
2-Nitropentachlor-1,3-butadien: Aus 2H-Pentachlorbutadien und 58
proz. HNO3 nach Lit.20 Ausb. 45% Sdp1 69–71◦C, n2D0: 1.5585 (Lit.20
43%, Sdp1 68–70◦C, n2D0:1.5583.)
1,3,4,4-Tetrachlor-1-(4-fluorphenylthio)-2-nitro-
1,3-butadien (3)
Zu 2.0 g (7.32 mmol) 1 werden bei Raumtemp. 0.47 g (3.66 mmol)
p-Fluorthiophenol gegeben und 36 h geru¨hrt. Die nach Zugabe von
Methanol entstandenen Kristalle werden mit Methanol gewaschen.
Ausb. 1.02 g (38%) gelbe, gla¨nzende Kristalle vom Smp. 99–100◦C (aus
Methanol).
–IR (KBr): ν = 3100 cm−1 (C H), 1595, 1605 (C C), 1300, 1540
(NO2). -1H-NMR (CDCl3, TMS int.): δ = 7.1–7.6 ppm (m, 4H, Aromaten-
H). C10H4SCl4NO2F (363.02). –MS:Molmasse: 363.0.
3,4,4-Trichlor-1,1-bis(4-fluorphenylthio)-2-nitro-
1,3-butadien (4)
Zu 2.0 g (7.32 mmol) 1 werden in wa¨βrigem Ethanol in Gegenwart
von Natronlauge bei Raumtemp. 0.94 g (7.32 mmo) p-fluorthiophenol
gegeben und 24 h geru¨hrt. Die nach Zugabe von Methanol entstandenen
Kristalle werden mit Methanol gewaschen. Ausb. 0.211 g (60%) gelbe,
gla¨nzende Kristalle vom Smp. 88–90◦C (aus Methanol).
–IR (KBr): ν = 3050, 3100 cm−1 (C H), 1590 (C C), 1350,
1540 (NO2). -1H-NMR (CDCl3, TMS int.): δ = 6.5–7.8 ppm (m, 8H,
Aromaten-H). C16H8S2Cl3NO2F2 (454.732). –MS:Molmasse: (454.8).:

3,4-Dichlor-1,1,4-tris(4-fluorphenylthio)-2-nitro-
1,3-butadien (5)
Zu 0.1 g (0.27 mmol) 3 werden in wa¨βrigem Ethanol in Gegen-
wart von Natronlauge bei Raumtemp. 0.035 g (7.32 mmol) p-
Fluorthiophenol gegeben und 24 h geru¨hrt. Die nach Zugabe von
Methanol entstandenen Kristalle werden mit Methanol gewaschen.
Ausb. 0.045 g (30%) gelbe, gla¨nzende Kristalle vom Smp. 75–76◦C (aus
Methanol).

Allgemeine Arbeitsvorschrift (AAV) fu¨ r die Umsetzung von
Mono(arylthio)-2-nitrobutadien (3) mit den Aminen (6, 8, 10,
12, 14)
Zu 0.1 g (0.27 mmol) 3 werden in Ether bei Raumtemp. die jeweils
angegebenen Mengen Amin dazugegeben und 24 h geru¨hrt. Nach Zu-
gabe von 100 ml Wasser wird mit Chloroform extrahiert, die Chloro-
formphase mit Wasser gewaschen, mit MgSO4 getrocknet und i.Vak.
¨
eingedampft. Das erhaltene gelbe Ol wird kristallisiert.
3,4,4-Trichlor-1-(4-fluorphenylthio)-1-(4-morpholino)-
2-nitro-1,3-butadien (7)
Dargestellt nach der AAV aus 0.1 g (0.27 mmol) 3 und 0.024 g
(0.27 mmol) Morpholin 6.
