tumors. Finally, we studied the anti-proliferation activity of Notes and references
PNF on A549R tumor spheroids with a diameter of approxi-
mately 400 mm. After incubation with PNF, we found that PNF
obviously penetrated into the spheres (Fig. S17, ESI†). After

day of incubation with PNF (20 mM), the spheroids began to

loosen, whereas the spheroids treated with cisplatin (40 mM)
did not begin to loosen until the third day (Fig. S18, ESI†). In
contrast, the control spheroids remained almost unchanged
after 8 days of incubation. These results indicate that PNF
exhibits more potential anti-proliferative activity than cisplatin,
thereby affecting the integrity of the MCTSs more rapidly and
effectively. In addition, the status of cancer cells in MCTSs after
PNF treatment was assessed using double staining assay
with acridine orange (AO, live cells, green fluorescence) and
ethidium bromide (EB, dead cells, red fluorescence) (Fig. 3c).
We found that PNF had a great potential to induce cancer cell
death in spheroids and to significantly reduce the viability of
live cancer cells. Therefore, PNF could effectively kill tumor
cells and disrupt the structure of tumor spheroids.

In summary, we have developed PNF, a fluorogenic small
molecule-based platform that contains an H S-induced reduc-

tive azide group, which can be activated by overexpressed H S
1

in the cancerous environment. This activation serves to give a
significant turn-on fluorescence response and release the
potent anticancer drug, amonafide (ANF). The introduction of
the azide group significantly improved the lipophilicity and
biocompatibility of PNF, which could be effectively uptaken by
live A549R cancer cells and mainly targeted in lysosomes.
Surprisingly, PNF can be activated and release ANF in lyso-
somes, from which it then escapes into the nucleus. The
cytotoxicity mechanisms include DNA damage, cell cycle arrest
in the G2/M phase, and mitochondrial disruption, eventually
resulting in the autophagy of cancer cells. Importantly, PNF
