Publisher SummaryThis chapter discusses the synthesis, special properties, microbial kinetics, pharmacology, and structural-activity relationship of lincomycin hydrochloride. Lincomycin hydrochloride monohydrate is a white or almost white, crystalline powder. It is odorless or has a faint mercaptan - like odor and has a bitter taste. Different methods were developed to synthesize lincomycin. Most of these methods directed towards the synthesis of 6-amino-6,8-dideoxy-D-erythro-D-galactose commonly named lincosamine. The D – galactose-derived aldehyde is the most frequently used synthon. Other direction were concentrated on the synthesis of lincomycin from non-carbohydrate precursors through the application of furan compound or heteroDiels—Alder Approach. Lincomycin hydrochloride has two polymorphic forms( A and B ) . This is detected by their x-ray diffraction, which showed the following interplanar spacing for the two crystalline forms. The mass spectrum of lincomycin showed five characteristic and diagnostic ions (molecular ion and four other ions) which can be considered to arise from molecular ions with charge localization at either the amino nitrogen or the pyran oxygen. These ions provided considerable insight into the structure elucidation of lincomycin. Microbial kinetic studies were applied to the action of lincomycin and clindamycin against Staphylococcus aureus, a representative gram-positive Coccus. It is found that the effective ratio of a clindamycin-lincomycin combination against Staphylococcus aureus is ranged between 6:1 and 9:1.
