The measurement of serum/plasma levels of 1α,25-dihydroxyvitamin D₃ 1 a is important for the diagnosis influencing vitamin D metabolism.To obtain antibodies to the metabolite 1a which are highly specific and useful for development of immunoassays, two novel haptenic derivatives, 11α-(3-carboxypropanoyloxy)-1α,25-dihydroxyvitamin D₃ 2a and 11α-(4-carboxybutanoyloxy)-1α,25-dihydroxyvitamin D₃ 2b were synthesized each in 19 steps from a suitably protected derivative (3) of 11α,25-dihydroxycholesterol.
