leading to entA-1a) to give the desired urea ent-1l. Analogous
synthetic routes led to other analogs shown in Tables 1 and 2.
In conclusion, we have identified a novel spirocyclic amine-de-
rived urea class as potent and selective sEH inhibitors with good PK
and ex vivo target engagement. However, rac-1a, a representative
compound of this series, failed to elicit a significant blood pressure
lowering effect in SHRs. More structurally diverse sEH inhibitors
will be reported in due course.
