Forced degradation of L-(+)-bornesitol, a bioactive marker of Hancornia speciosa: Development and validation of stability indicating UHPLC-MS method and effect of degraded products on ACE inhibition
The antihypertensive activity of the medicinal plant Hancornia speciosa has been previously demonstrated by us, being the activity ascribed to polyphenols and cyclitols like L-(+)-bornesitol. We herein evaluated the stability of the bioactive marker bornesitol submitted to forced degradation conditions. Bornesitol employed in the study was isolated from H. speciosa leaves. An UHPLC-ESI-MS/MS method was developed to investigate bornesitol stability based on MRM (Multiple Reaction Monitoring) acquisition mode and negative ionization mode, employing both specific (m/z 193 → 161 Da) and confirmatory (m/z 193 → 175 Da) transitions. A gradient elution of 0.1% formic acid in water and acetonitrile was performed on a HILIC column. The method was validated and showed adequate linearity (r2 > 0.99), selectivity, specificity, accuracy, and precision (RSD 2.9%). The method was robust for deliberate variations on dessolvation temperature, but not for changes in the flow rate and dessolvation gas. The results from the stability studies allowed us to classify bornesitol as labile for acidic and alkaline hydrolysis, but as very stable for oxidative and neutral hydrolysis exposure. Bornesitol was categorized as practically stable under photolysis degradation, whereas a considerable reduction on its contents was induced by metal ions and thermolysis exposure. Degraded samples from neutral hydrolysis and thermolysis were assayed in vitro for ACE inhibition and showed a substantial decrease in biological activity as compared to intact bornesitol. myo-Inositol was identified as the major degradation products in both matrices. This is the first report on bornesitol stability under different stress conditions and the obtained data are relevant for the development and quality control of standardized products from H. speciosa leaves.

To elucidate the biological functions of these molecules, robust techniques to characterize inositol phosphate metabolism at the cellular level are highly sought after. Method for the production of inositol phytin
The invention discloses a method for preparing inositol by adopting phytine. The method comprises the following steps: with the phytine as a raw material, adding deionized water and the phytine into a reactor according to a weight ratio of (5-20):1 of liquid to material, regulating the pH value to 4-5.5, adding phytase of which the weight is 0.5-3 percent of that of the phytine, stirring for hydrolysis under the assistance of ultrasonic waves, and controlling the temperature of a reaction system to be 37-50 DEG C, wherein the action time of the ultrasonic waves is 0.5-1 hour in the hydrolysis process, and the start time of the action of the ultrasonic waves is 0-50 percent of the total reaction time; ending the hydrolysis reaction after the hydrolysis is performed for 4-24 hours, adding solid super-acid of which the weight is 0.2-1 percent of that of the phytine into hydrolysate, performing complete hydrolysis for 15-45 minutes under a pressure of 0.12-0.20 MPa at the reaction temperature of 105-120 DEG C, filtering, adding filtrate into a water solution of calcium oxide, regulating the pH to 7.0, filtering, concentrating the filtrate until the ratio of the liquid to the material is 2:1 based on the initial weight of the phytine, then adding ethanol in a plurality of steps and respectively separating and precipitating to finally obtain the inositol. The method disclosed by the invention has the advantages of low pressure, low temperature and low energy consumption.

Elaboration of the ether cleaving ability and selectivity of the classical Pearlman's catalyst [Pd(OH)2/C]: Concise synthesis of a precursor for a myo-inositol pyrophosphate
The cleavage of propargyl, allyl, benzyl, and PMB ethers by Pd(OH) 2/C can be tuned in that order, by varying the reaction conditions. Other moieties such as C-C double bonds, esters, trityl ether, p-bromo and p-nitrobenzyl ethers are stable to these reaction conditions. Cleavage of allyl ethers can be made catalytic by using 1:1 mixture of Pd(OH)2/C and Pd/C. The synthetic potential of the selective ether cleaving ability of Pd(OH)2/C, essentially under neutral conditions, has been demonstrated by an efficient synthesis of a precursor for the preparation of an inositol pyrophosphate derivative.

In vitro and in vivo antioxidant activity of a water-soluble polysaccharide from dendrobium denneanum
The water-soluble crude polysaccharide (DDP) obtained from the aqueous extracts of the stem of Dendrobium denneanum through hot water extraction followed by ethanol precipitation, was found to have an average molecular weight (Mw) of about 484.7 kDa. Monosaccharide analysis revealed that DDP was composed of arabinose, xylose, mannose, glucose and galactose in a molar ratio of 1.00:2.66:8.92:34.20:10.16. The investigation of antioxidant activity both in vitro and in vivo showed that DDP is a potential antioxidant.

Conformational study of the natural iron chelator myo-inositol 1,2,3-trisphosphate using restrained/flexible analogues and computational analysis
Myo-Inositol 1,2,3-trisphosphate [Ins(1,2,3)P3], a component in mammalian cells, possesses the correct chemical properties of an intracellular iron transit ligand. Here we have examined the conformation of the Ins(1,2,3)P3-Fe3+ complex. The synthesis and antioxidant properties of 4,6-carbonate-myo-inositol 1,2,3,5-tetrakisphosphate [4,6-carbonate Ins(1,2,3,5)P4], which is locked in the unstable penta-axial chair conformation and 1,2,3-trisphosphoglycerol, a flexible acyclic analogue of Ins(1,2,3)P3, are reported. 4,6-Carbonate Ins(1,2,3,5)P4 caused complete inhibition of iron-catalysed hydroxyl radical (HO?) formation at 100 μM, thereby resembling Ins(1,2,3)P3 and supporting a penta-axial chair binding conformation. In contrast, 1,2,3-trisphosphoglycerol was shown to have incomplete antioxidant properties. In support of experimental observations, we have applied high-level density functional calculations to the binding of Ins(1,2,3)P3 to iron. This study provides evidence that Fe3+ binds tightly to the less stable penta-axial conformation of Ins(1,2,3)P3 using terminal and bridging phosphate oxygens, thought to also contain a tightly bound water molecule or hydroxyl ligand in the complex.

BIOOCOMPATIBLE POLYMER COMPOSITIONS
The present invention provides a biocompatible prepolymer comprising hydrophilic and hydrophobic segments, wherein the hydrophobic segments have at least one ethylenically unsaturated functional group and at least 5% of the segments have two or more ethylenically unsaturated functional groups and water. The invention further provides a biocompatible prepolymer composition comprising hydrophilic and hydrophobic prepolymers, wherein at least one of the hydrophobic prepolymers has at least one ethylenically unsaturated functional group and at least 5% of the prepolymers have two or more ethylenically unsaturated functional groups and water. The invention further provides use of the prepolymer or prepolymer compositions of the invention in biomedical applications such as tissue engineering, as bone substitutes or scaffolds, and in wound treatment.

Kinetic characterization of O-phospho-L-tyrosine phosphohydrolase activity of two fungal phytases
Fungal phytases belonging to "histidine acid phosphatase" or HAP class of phosphohydrolases that catalyze the hydrolysis of phytic acid could also hydrolyze O-phospho-L-tyrosine, which is also called phosphotyrosine. Two phytases from Aspergillus niger and Aspergillus awamori with pH optima 2.5 were tested for phosphotyrosine hydrolase activity; both enzymes cleaved the phosphomonoester bond of phosphotyrosine efficiently at acidic pH. The K m for phosphotyrosine ranged from 465 to 590 μM as opposed to 135 to 160 μM for phytate. The Vmax, however, is 2-4 times higher for phosphotyrosine than it is for phytate. The catalytic efficiency of phytase for phosphotyrosine is on the same order as it is for phytate (3.5 × 10 6 to 1.6 × 107 M-1 s-1); the pH versus activity profile for phosphotyrosine is, however, different from what it is for phytate. The temperature optima shifted 5°C higher to 70°C when phosphotyrosine was used as the substrate. Taken together, the kinetic data show that fungal HAPs that are known as PhyB are capable of cleaving the phosphomonoester bond in phosphotyrosine. This is the first time that phosphotyrosine phosphatase (PTPase) activity has been reported for the subgroup of HAP known as phytase.

Hydroxyl group deprotection reactions with Pd(OH)2/C: a convenient alternative to hydrogenolysis of benzyl ethers and acid hydrolysis of ketals
Benzyl ethers, ketals and orthoformates were cleaved with Pd(OH)2/C in methanol, to generate the corresponding alcohol; carboxylic acid esters were stable under these reaction conditions. Pd(OH)2/C in methanol was used for the deprotection of hydroxyl groups during the preparation of sequoyitol via myo-inositol orthobenzoate. This method of deprotection has the potential to be useful in the synthesis of different classes of organic compounds since the reaction conditions do not involve strong acids, bases or hydrogen.

Stereochemistry at phosphorus of the reaction catalyzed by myo-inositol monophosphatase
myo-Inositol monophosphatase (IMPase), the proposed target for lithium therapy for manic depression, is an important enzyme in the biosynthesis of second messengers. Earlier studies have shown that the IMPase-catalyzed hydrolysis of myo-inositol monophosphates to inorganic phosphate and myo-inositol proceeds by direct attack of water at phosphorus. However, research groups have independently proposed either an in-line displacement (with inversion of stereochemistry at phosphorus) or an adjacent attack with a pseudorotation (with retention of stereochemistry at phosphorus). Here, the elucidation of the stereochemical pathway is presented. The IMPase-catalyzed hydrolysis of D-1-Sp-myo-inositol [17O]-thiophosphate in the presence of H218O gave inorganic Rp-[16O, 17O, 18O]-thiophosphate, with inversion of configuration at phosphorus. This is only consistent with an in-line displacement, and it rules out the controversial adjacent/pseudorotation mechanism. This result will assist in the design of alternative inhibitors of IMPase.

Novel synthesis of enantiomerically pure natural inositols and their diastereoisomers
The various inositol polyphosphates have been found to trigger many important biological processes. Although the knowledge of this phosphoinositide signaling system has been discovered in the past 10 years, many factors remain unclear. For this reason, there is an increased demand for supplies of D-myo-inositol and particularly of novel analogues to investigate these biological mechanisms in more detail. Herein, we report the efficient syntheses of all diastereoisomers of inositol starting with 6-O-acetyl-5-enopyranosides. Conversion of 6-O-acetyl-5-enopyranosides into the corresponding substituted cyclohexanones (Ferrier-II rearrangement) was found to proceed efficiently with a catalytic amount of palladium dichloride. Stereoselective reduction of β-hydroxy ketones obtained provided the precursors to all inositol diastereoisomers in good to excellent yields and with high stereoselectivities. Good accessibility of these enantiomerically pure inositol diastereoisomers results in the efficient syntheses of D-myo-inositol 1,4,5-trisphosphate and D-myo-inositol 1,3,4,5-tetrakisphosphate.

13C Labelling and electrospray mass spectrometry reveal a de novo route for inositol biosynthesis in Leishmania donovani parasite
In this paper we provide evidence for the presence of a de now route for myo-inositol biosynthesis in the protozoan parasite Leishmania donovani, by the application of 13C labelling and electrospray ionisation mass spectrometry (ESMS). For this myo-[1-13C]inositol is prepared from D-[6-13C]glucose and biosynthetically incorporated in the parasite promastigote cell culture. Biosynthetic phosphatidylinositol (PI) and its hydrolysis products glycero-PI and inositol are analysed by ESMS and the isotopomeric ratio determined. The incorporation experiments show substantial isotopic dilution, indicating the presence of myo-inositol 1-phosphate synthase (MIP synthase) enzyme in the parasite; this is further confirmed by incorporation of D-[6-13C]glucose in the parasite phosphatidylinositol. The pathway of dephosphorylation of myo-inositol hexakisphosphate by phytases from wheat bran of Triticum aestivum L. cv. Nourin #61
Phytases are the primary enzymes responsible for the hydrolysis of phytic acid, myo-inositol-1, 2, 3, 4, 5, 6-hexakisphosphate (InsP6). The pathway of hydrolysis of InsP6 by phytase from wheat bran of Triticum aestivum L. cv. Nourin #61 is proved in this study. Structures of the intermediates were established by a variety of nuclear magnetic resonance techniques (1H-, two-dimensional 1H-1H coupling-correlation spectra and two-dimensional 31P-1H correlation spectra), gas chromatography, and bioassay. On the basis of the structures identified, initial hydrolysis of the phosphate ester occurs at the D/L-4 position of InsP6 to yield D/L-Ins (1, 2, 3, 5, 6) P5. After the dephosphorylation, the pathway of dephosphorylation is divided into two routes. The main route proceeds via D/L-Ins (1, 2, 5, 6) P4, D/L-Ins (1, 2, 6) P3 and D/L-Ins (1, 2) P2, while the minor route proceeds via D/L-Ins (1, 2, 3, 6) P4, Ins (1, 2, 3) P3 and D/L-Ins (1, 2) P2. D/L-Ins (1, 2) P2 is hydrolyzed at the D/L-1 or 2-position, and finally myo-inositol is produced.

Process for recovering phytic acid, lactic acid or inositol
Described is a process for recovering phytic acid or phytic acid salt from a medium in which it is contained. This process includes contacting the medium with a solid-phase free base polymer having tertiary amine functions to adsorb the phytic acid or salt thereof. In one preferred mode, the medium to be treated also contains lactic acid, and separate fractions predominant in (i) lactic acid and (ii) phytic acid or phytic acid salt, respectively, are recovered. Perfluoro-1H,-1H-neopentyl containing contrast agents and method to use same
Organic compounds for diagnostic imaging which contain at least one aryl group which has been derivatized to contain at least one perfluoro-1H,1H-neopentyl moiety are provided. The perfluoro-1H,1H-neopentyl groups produce a single magnetic resonance to insure a maximum signal to noise ratio. This paper reports on the details of a general design of carbohydrates and cyclitols from biocatalytically derived synthons.Homochiral 1-halogenocyclohexa-4,6-diene-2,3-diols 1a and 1b have been generated from chloro- and bromobenzene, respectively, by means of bacterial dioxygenase of Pseudomonas putida 39D.These chiral synthons have been manipulated to cyclitols and carbohydrates by further stereoselective functionalizations.The preperation of D-chiro-inositol, neo-inositol, muco-inositol, and allo-inositol exemplifies their use in enantiocontrolled synthesis.A novel oxidation of polarized dienes with KMnO4 resulted in the synthesis of α-halogeno epoxy diols, which proved unexpectedly stable.A mechanism is proposed for this transformation and placed in context with the only four reported examples of this reaction in the literature.In addition to the application of this new chemistry to the synthesis of cyclitols, chloro epoxy diol 21a has been transformed into a series of cyclitol synthons by reductive or hydrolytic operations.Reaction of 21a with ammonia led to the preparation of highly oxygenated pyrazines, whose structure were proven by X-ray crystallography.The use of 21a in the preparation of D-chiro-3-inosose, a hitherto unreported cyclitol derivative, is also reported.In addition, chloro epoxy diol 21a was transformed into D-erythruronolactone, completing the synthesis of this important chiral pool reagent in two operations from chlorobenzene.Oxidative cleavage of tetrol 20 yielded D-mannosolactone identical with an authentic sample.

The disaccharides related to glycosyl phosphatidyl inositol anchors of membrane proteins, 4-O-(2-amino-2-deoxy-α-D-glucopyranosyl)-D-myo-inositol-1-phosphate and 4-O-(2-amino-2-deoxy-α-D-galactopyranosyl)-D-chiro-inositol-1-phosphate, have been prepared from optically resolved myo-inositol derivatives.The chiro-inositol moiety was obtained by epimerization of a selectively blocked myo-inositol-triflate ester.The resolved inositols were subsequently phosphorylated to yield the disaccharide aglycones.The amino-sugar components were prepared by azidonitration of suitably protected glucal and galactal derivatives.The derived pyranosyl chlorides were then condensed with the inositol phosphates using silver triflate as the promoter.

Myo-inositol Polyphosphate Intermediates in the Dephosphorylation of Phytic Acid by Acid Phosphatase with Phytase Activity from Rice Bran
Myo-inositol phosphate intermediates formed during desphosphorylation of phytic acid (by rice bran acid phosphatase with phytase activity F2) were isolated by ion-exchange chromatography.Hydrolysis of phytic acid proceeded via L-myo-inositol 1,2,3,4,5 and D-myo-inositol 1,2,4,5,6-pentakisphosphate, D-myo-inositol 1,2,5,6 and L-myo-inositol 1,2,3,4-tetrakisphosphate, D-myo-inositol 1,2,6 and myo-inositol 1,2,3-trisphosphate, D-myo-inositol 1,2-bisphosphate and finally myo-inositol monophosphate (myo-inositol 2, 93percent; D-myo-inositol 1, 7percent).Six percent of myo-inositol and 85percent of Pi were found in the reaction mixture when the hydrolyses of phytic acid by the enzym did not proceed further.The initial points of dephorylation of phytic acid by the phytase took place at L-myo-inositol 6 (85percent), D-myo-inositol 3 (7percent), D-myo-inositol 2 (5percent), and myo-inositol 5 (3percent) at the positions of the myo-inositol ring.

Polyfunctionalised Cyclohexanes from Dianhydroinositols. cis-1,3(1,4)-Inosadiamines from Benzene
The preparative value of the dianhydroinositols 4 - 6, which are readily available from benzene, for the total synthesis of cis-1,4- and cis-1,3-disubstituted cyclohexanetetrols is demonstrated.With monovalent reagents (H2O, HI, NaN3, NH2NH2) the twofold epoxide opening proceeds regioselectively (1,4-disubstitution) yielding the muco- and chiro-cyclohexane derivatives 9, 17, and 26.With hydrazine and N,N'-dimethylhydrazine as 1,2-dinucleophiles the cis-1,3-inosadiamines 7 g (5-epistreptamine) and 16 g, j (2-epistreptamine, actinamine) are obtained from 4/5 in good yields (75 - 90percent).From the less symmetrical 6, however, only the mixture of 25 g (streptamine) and 27 g is produced in modest yield (ca. 50percent).The epoxide opening reactions starting from 4 - 6 are kinetically so similar that a selective opening is not possible.

Preparation of antitumor agent from shellfish
A new antitumor agent is provided which consists essentially of water-soluble, macromolecular glycoprotein substances having a range of molecular weights within the limits of from 10,000 to 300,000 and which is prepared from shellfish or mollusc by taking up the liquid portion which comes from the cooking of raw shellfish in an aqueous solvent, typically water or a saline solution, or vapor of such solvent and which is to be discarded as waste, concentrating the liquid portion to a smaller volume or to dryness, dissolving the resulting concentrate or dry powder in an aqueous solvent and isolating the desired water-soluble, macromolecular glycoprotein substances therefrom. The isolation of the desired substances is preferably carried out by an ion-exchange chromatography with a basic anion-exchanger followed by a gel-filtration.

PURIFICATION AND STRUCTURE DETERMINATION OF THREE α-D-GALACTOPYRANOSYLCYCLITOLS FROM SOYA BEAN
