The protection of the hydroxy group of p-cresol 1 by o-silylation gives
derivatives 2 and 3, the methyl group of which can be brominated by
NBS. The phase transfer catalysis applied to 4 and 5 is a good way
which permits the mild introduction of the allylthio group (6 and 7).
Hydroboration applied to silylated compounds 8 and 9, followed by
methanolysis and hydrolysis leads to target acids 10 and 11 in a good
yield.
La protection du groupement hydroxy du p-cre´sol 1 par o-silylation
donne les de´rive´s 2 et 3 ce qui permet de bromer le substituant me´thyle
par le N-bromosuccinimide (NBS). La catalyse par transfert de phase
(CTP) applique´e aux produits 4 et 5 est une bonne me´thode pour in-
troduire un groupement allylthio (compose´s 6, 7). L’hydroboration des
compose´s silyle´s 8 et 9, suivie d’une me´thanolyse et d’une hydrolyse
permet d’acce´der aux acides cibles 10 et 11 avec de bons rendements.

Les de´rives organobore´s pre´sentent un grand inte´reˆt dans le domaine
de la synthe`se organique.1−3 Nous nous sommes inte´resse´s aux acides
γ -thio-boroniques 10 et 11, compose´s susceptibles de jouer un roˆle de
me´diateur dans le transport transmembranaire a` cause de leur ressem-
blance avec les acides γ -amino-boroniques.4
´
RESULTATS ET DISCUSSION
Pour atteindre les compose´s 10 et 11, nous avons choisi d’hydroborer
les allyl-sulfures correspondants obtenus a` partir du p-cre´sol.

1) Protection, Ethoxyalkylation du p-cre´ sol
Le traitement du p-cre´sol 1 par l’hexame´thyldisilazane (HMDS) ou la
chlorhydrine silyle´e Cl-(CH2-CH2-O)2)-SiMe3 1a donne respectivement
les de´rive´s silyle´s 2 et 3 (Sche´ma 1). Le de´rive´ 2 est aise´ment obtenu de
manie`re quantitative et sa de´protection se fait par simple hydrolyse.5−6
En contre partie, la fragilite´ de cette protection nous oblige a` manipuler
ces compose´s sous argon. Les motifs e´thoxyle´s du de´rive´ 3 sont destine´s
a` accroˆıtre l’hydrosolubilite´ de l’acide boronique cible.

2) Bromation Radicalaire et Introduction du
Groupement Allylthio
La bromation radicalaire7 du groupe me´thyle conduit aux de´rive´s 4 et
5 (e´tape 1 du Sche´ma 2. Nous avons utilise´ la CTP8−9 en pre´sence de
chlorure de trie´thylbenzylammonium (TEBA) comme agent de trans-
fert de phase pour la substitution du brome par le groupe allylthio.
Cette me´thode nous a permis d’acce´der aux produits 6 et 7 en e´vitant
les re´actions e´quilibre´es et en ame´liorant le temps de la re´action.
Cependant on a remarque´ la disparition du groupement protecteur
trime´thylsilyle (e´tape 2 du Sche´ma 2).

3) Hydroboration, Me´ thanolyse et Hydrolyse Acide
La de´silylation observe´e a` l’e´tape pre´ce´dente ne´cessite une nouvelle
silylation des compose´s 6 et 7 pour donner les de´rive´s 8 et 9.
Les conditions standards d’hydroboration10 applique´es aux produits
silyle´s 8 et 9 nous conduit apre`s me´thanolyse et hydrolyse acide aux
acides 10 et 11 avec un de´gagement de deux e´quivalents d’hydroge´ne ce
qui montre que deux boranes interme´diaires non isole´s 8a et 9a ont e´te´

forme´s. La RMN H montre la disparition du groupement protecteur
trime´thylsilyle (Sche´ma 3).

PARTIE EXPERIMENTALE ET CARACTERISATION
Les spectres RMN 1H ont e´te´ enregistre´s avec un spectrome`tre Bru¨ker
AC-250. Les de´placements chimiques sont exprime´s en partie par mil-
lion (δ ppm) par rapport au chloroforme deute´re´. Les abre´viations suiv-
antes sont utilise´es: s pour singulet, d pour doublet, dd pour doublet de
doublet, t pour triplet et m pour multiplet.
[2-(2-p-tolyloxy-e´ thoxy)-e´ thoxy)]-trime´ thylsilane (3)
Dans un tricol de 100 ml, place´ dans un bain de glace et sous atmosphe`re
d’argon et contenant 50 ml d’e´ther sont ajoute´s 10 mmol d’hydrure de
sodium, puis tre`s lentement 10 mmol de p-cre´sol. La re´action est suivie
par la mesure du de´gagement d’hydroge`ne.
Lorsque celui-ci est termine´, 10 mmol de chlorhydrine silyle´e sont
ajoute´es goutte a` goutte.
Le milieu re´actionnel est ensuite laisse´ sous agitation pendant deux
heures. On distille sous pression re´duite.

La chlorhydrine silyle´e (1a) est pre´pare´e par action du HMDS sur le
2-(2-chloroe´thoxy) e´thanol suivant le protocole de´crit ci-dessous.
Aspect: liquide incolore, Rdt: 90%, Eb: 132◦C/1 mmHg.
RMN 1H (CDCl3) δH: 0.05 (s; 9H; SiMe3), 2.1 (s; 3H; CH3), 3.64–4.11
(m; 8H; (CH2 CH2 O)2 ), 6.8–7.1 (m; 4H; Ph).
Protection par HMDS des Compose´ s Hydroxyle´ s
Dans un tricol muni d’un re´frige´rant et e´quipe´ d’une agitation
magne´tique on introduit 0.1 mol de produit hydroxyle´ et 0.15 mol
d’he´xame´thyldisilazane. Le me´lange est porte´ a` reflux pendant 12
heures sous argon. Les produits obtenus sont purifie´s par distillation
sous pression re´duite et stocke´s sous argon.

2-(2-chloroe´ thoxy) Ethoxytrime´ thylsilane (1a)
Aspect: liquide incolore; Rdt: 90%; Eb: 80◦C/1 mmHg.
RMN 1H (CDCl3) δH: 0.05 (s; 9H; SiMe3), (3.5–3.7 (m; 8H;
(CH2 CH2 O)2 ).
(4-Me´ thyl-phe´ noxy)-trime´ thylsilane (2)
Aspect: liquide incolore, Rdt: 95%, Eb: 76◦C/1 mmHg.

(m; 4H; Ph).
(4-Allylsulfanylme´ thyl-phe´ noxy)-trime´ thylsilane (8)
Aspect: liquide orange´, Rdt: 90%; Eb: 132◦C/1 mmHg.

Bromation Radicalaire
Dans un tricol, muni d’un re´frige´rant et e´quipe´ d’une agitation
magne´tique on place 75 mmol du compose´ silyle´ et 1.1 e´quivalent de
NBS dans 150 ml de CCl4. On ajoute une pointe de spatule d’AIBN (α,αꢁ-
azo-iso-butyronitrile) comme amorceur radicalaire. Le tout est prote´ a`
reflux sous agitation pendant 5 heures. On filtre a` chaud le succin-
imide forme´ au cours de la re´action. Le filtrat est e´vapore´ sous pression
re´duite.
(4-Bromomethyl-phenoxy)-trime´ thylsilane (4)
Aspect liquide jaune, Rendement quantitatif.
RMN 1H(CDCl3) δH; 0.05 (s; 9H; SiMe3), 4.45 (s; 2H; CH2 Br), 6.8–
7.1 (m; 4H; Ph).
{2-[2-(4-bromoe´ thylphe´ noxy)-e´ thoxy]-e´ thoxy}-
trime´ thylsilane (5)
Aspect: liquide jaune, Rendement quantitatif.
RMN 1H(CDCl3) δH: 0.05 (s; 9H; SiMe3), 4.46 (s; 2H; CH2 Br), 3.64–
4.11 (m; 8H; (CH2 CH2O)2), 6.83–7.30 (m; 4H; Ph).
Substitution nucle´ ophile
Dans un tricol, muni d’un re´frige´rant et d’une agitation magne´tique on
place 15 mmol du produit brome´ 4 ou 5, 50 ml de CCl4 et 0.75 mmol de
chlorure de benzyltributylammonium (TBBA) pur a` 98%.
On ajoute 16.5 mmol de d’allylthiolate de potassium dissous dans 3
a` 5 ml d’eau. Le tout est porte´ a` reflux sous forte agitation pendant
5 heures.
Apre`s retour a` la tempe´rature ambiante, la phase organique est lave´e
avec 15 a` 20 ml d’eau, se´che´e sur du sulfate de sodium, filtre´e et le
solvant est e´vapore´ sous pression re´duite.
L’allylthiolate de potassium est pre´pare´ en faisant re´agir 1
e´quivalent d’allylthiol a` 70% sur 1.4 e´q. de KOH dans 5 ml d’eau.
4-Allylsulfanylme´ thyl-phe´ nol (6)
Aspect: liquide visqueux marron, Rdt: 85%.
RMN 1H(CDCl3) δH: 3.02 (d; 2H; S CH2 Ph), 5–6 (m; 3H;
CH2 CH), 6.79–7.17 (m; 4H; Ph).

2-[2-(4-allylsulfanylme´ thyl-phe´ noxy)-e´ thoxy]-e´ thanol (7)
Aspect: liquide orange´, Rdt: 80%.
RMN 1H(CDCl3) δH: 3.02 (d; 2H; S CH2 CH CH2), 3.6 (s; 2H;
S CH2 Ph), 5–6 (m; 3H; CH2 CH), 6.79–7.17 (m; 4H; Ph).
Hydroboration
a` 10 mmol du compose´ 8 ou 9 dissous dans 50 ml de CCl4 maintenu a` 0◦C
par un bain de glace on ajoute 10 mmol de complexe-borane sulfure de
me´thyle(BMS). La re´action est agite´e a` tempe´rature ambiante pendant
3 heures.
Me´ thanolyse et hydrolyse
Lorsque l’hydroboration est finie l’appareillage est ensuite connecte´
a` un gazome´tre afin de suivre le de´gagement gazeux lors de la
me´thanolyse. Celleci est faite en ajoutant du me´thanol par portions a` la
solution jusqu’a` ce que le de´gagement d’hydroge´ne s’arreˆte. L’hydrolyse
acide est faite en ajoutant 5 ml d’HCL (2N) au me´lange re´actionnel. On
rassemble les phases organiques et le solvant est e´vapore´ sous pression
re´duite.
Acide 3-(4-hydroxybenzylsulfanyl) Propylboronique (10)
Aspect: liquide orange´, Rdt: 70%.
RMN 1H(CDCl3) δH: 0.9 (t; 2H; CH2 B), 1.61 (m; 2H;
CH2 CH2 CH2), 2.38 (t; 2H; S CH2 CH2), 3.62 (s; 2H; S CH2 Ph),
6.79–7.17 (m; 4H; noyau aromatique).
Acide 3-{4-[2-(2-hydroxye´ thoxy)-e´ thoxy]-benzylsulfanyl}
Propylboronique (11)
Aspect: liquide visqueux ambre´, Rdt: 80%.
