  Compounds 2–10 were assayed against established mammalian cell lines, including cancer cells, by measuring the inhibition of cell growth by an MTT assay (Table 1). Upon examination of the Tup position at the C-terminus of tubulysin D (2–5), we found that a wide range of modifi-  cations were tolerated (Table 1).
