Syntheses, absorption and fluorescence properties for a series of new enzyme substrates are described, which are derived from 7-hydroxycoumarins possessing electron-withdrawing substituents in position 3.The new substrates are advantageous over existing ones in that they exhibit longwave absorption and fluorescence maxima as well as large Stokes' shifts.In addition, their pKa values, which are usually between 6.0 and 7.0, allow the direct and continuous kinetic assay of hydrolases such as esterases, phosphatases, and sulfatases. - Keywords: Enzyme substrates; Absorption spectra; Fluorescence spectra
