A simple procedure is described for the preparation of modified guanosines via an oxidiazaphosphorine oxide guanosine derivative. This nucleoside has been successfully used in the synthesis of guanosine containing -(CH₂)₂OH, -(CH₂)₆NH₂, -NHNH₂ "handles", or a tethered acridine moiety at the 2-position.
