mixture was then cooled and the solid which separated was filtered off
and washed with dry ether to afford the title compound (3.65 g, 77%)
m.p. 203–204ꢀC (lit.[4] 203–205dꢀC).
Dimethyl 4-amino-2,3-bismethoxycarbonylthiophene hydrochloride: 2,3-
Bismethoxycarbonyl-4-oxotetrahydrothiophene[7] (1.32 g), hydroxylamine
hydrochloride (0.5 g) and methanol (2.5 mL) were boiled together under
reflux for 1 h. At the end of this time the mixture was cooled whereupon
it solidified and was diluted with ethyl acetate. The compound was filtered
off and washed with ethyl acetate to give a white solid (0.79 g, 52%) m.p.
186–188dꢀC. A sample of this material afforded the yellow free base, m.p.
61.5–63.5ꢀC (lit.[7] m.p. 60–61ꢀC) on treatment with aqueous ammonia. An
analytical sample of the hydrochloride, crystallized from methanol/ether,
had m.p. 193–195dꢀC (Found: C, 38.1; H, 3.9; N, 5.6. C8H10ClNO4S
requires C, 38.2; H, 4.0; N, 5.6%).
4-Amino-3-methoxycarbonyl-2-methylthiophene
hydrochloride:
3-
Methoxycarbonyl-2-methyl-4-oxotetrahydrothiophene[5] (1.11 g), hydroxyl-
amine hydrochloride (0.5 g) and ethanol (2.5 mL) were stirred and boiled
under reflux in an oil bath at 100–105ꢀC for 1 h. The mixture solidified on
cooling; it was diluted with ethyl acetate and chilled in ice water before the
pale pink solid was filtered off and dried at the pump, the product weighted
0.65 g (49%) and had m.p. 183–185dꢀC (lit.[5] m.p. 179–181ꢀC).
