- Lysinol: A renewably resourced alternative to petrochemical polyamines and aminoalcohols

This paper reports the preparation of lysinol (2,6-diamino-1-hexanol) by the hydrogenation of lysine and an example of its use as a replacement for petrochemical-derived amines. Lysine is presently manufactured by fermentation of sugars and other carbon sources at scale exceeding 109 kg per year. Therefore, lysinol is potentially a renewable, platform aminoalcohol of previously unrecognized potential. Lysine hydrogenation proceeds under relatively modest conditions with Ru/C catalyst in water (100-150 °C, 48-70 bar, pH 1.5-2) to give lysinol in good yield (100% conversion, >90% selectivity; 50-70% isolated yield after purification by distillation). The impact of the various reaction parameters on conversion and selectivity are presented and discussed. Lysine hydrogenation at higher temperatures provides a pathway to piperidines and other products via further reduction and elimination of lysinol. The feasibility of lysinol synthesis from commodity, animal feed-grade lysine sources is presented as well. An example of the potential utility of lysinol is demonstrated by its use as a diamine curing agent with a standard epoxy resin. Novel piperidine and piperazine derivatives have been designed and tested as inhibitors of LTA4 hydrolase (LTA4H). Most potent compounds showed good potency in both enzymatic and functional human whole blood assay. Crystallography studies further confirmed observed structure-activity relationship and LTA4H binding mode for analogs from the piperidine series.

Ketimines, enamines and oxazolidines are moisture labile functional groups that in the presence of water undergo hydrolysis to yield free amines. Within the art such latent amines have found utility in curable compositions where it is desirable to initiate cure in the presence moisture. A number of novel polyketimines, polyenamines, polymeric oxazolidines and oxazolidines are disclosed. In particular, polymeric compounds of the above classes derived from C6 cyclic diketones and polyamines are reported. Suitable C6 cyclic diketones include cyclohexanediones and quinones. The invention further relates to the applications of the materials, such as in moisture cure adhesives.

Hydrogenation of pyridyl and quinolyl compounds 2-substituted with a carbonyl group (1a-c and 2b,c) using PtO2 and 1 equiv. of HCl (conditions A) provides clean and total formation of the desired amino alcohol (hydrogenation of the heterocyclic ring and of the carbonyl) while under conditions B1 and/or B2 (concentrated HCl or pure CF 3CO2H) the heterocyclic ring remains untouched and other aromatic parts are hydrogenated providing complex mixtures. When the heterocyclic ring is substituted by an alkyl group (quinaldine 3) conditions A provide mixtures while under conditions B2 (pure CF 3CO2H) the benzene ring is cleanly hydrogenated leading to a pure product.

Treating N-substituted pyrrolidines and piperidines bearing an allylic chain α to the nitrogen with strong bases leads to the opening of the heterocycle and provides 1,3-dienes disubstituted with an alkoxy and an aminoalkyl chain. The effects of the base and the solvent have been studied, as well as the influence of the ring size and the nitrogen substituent. The results obtained suggest a possible pre-chelation of the base cation before the deprotonation.

- Total synthesis of (+)-aloperine. Use of a nitrogen-bound silicon tether in an intramolecular diels-alder reaction

Enantioselective total syntheses of aloperine (1), N-methylaloperine (2), and N-allylaloperine (3) are reported. The central element of the synthetic strategy is an intramolecular Diels-Alder reaction in which the cycloaddends are tethered by a N-silylamine linkage. The total synthesis of 1 proceeds from commercially available 3-hydroxypiperidine hydrochloride (54) and (R)-pipecolinic acid (35) by way of nine isolated and purified intermediates. The synthesis is sufficiently efficient that gram quantities of (+)-aloperine (1) can be readily prepared. Early exploratory studies also introduced a convenient method for tethering cycloaddition partners with a sulfonamide unit to realize the intramolecular Diels-Alder cycloaddition of a vinylsulfonamide: 45 → 46.
